1.       TYPE OF DRUG- DAPSONE
Dapsone, also known as diaminodiphenyl sulfone (DDS)

 

2.       INDICATIONS (USE) – DAPSONE
This medication is an antibacterial drug, prescribed for leprosy and skin infections either alone or with other medications. It stops the bacterial growth.

 

3.       MECHANISM OF ACTION (MOA) – DAPSONE
Dapsone acts against bacteria and protozoa in the same way as sulphonamides, that is by inhibiting the synthesis of dihydrofolic acid through competition with para-amino-benzoate for the active site of dihydropteroate synthetase. The anti-inflammatory action of the drug is unrelated to its antibacterial action and is still not fully understood.

 

4.       ROUTES OF ADMINISTRATION- DAPSONE
Oral

 

5.       ONSET OF EFFECT OR ACTION- DAPSONE
Within 30-150 hrs.

 

6.      DOSAGE (DOSING INFORMATION) – DAPSONE
Adult: PO- The recommended dose is 50 – 300mg/day.

 

7.       HALF LIFE (DURATION OF ACTION) – DAPSONE
Half-life: 28 hr

 

8.       ADVERSE EFFECTS OR SIDE EFFECTS – DAPSONE
  • an allergic reaction (difficulty breathing; closing of the throat; swelling of the lips, tongue, or face; or hives);
  • bluish skin color;
  • muscle weakness;
  • numbness or tingling;
  • abdominal pain;

 

9.       CONTRAINDICATIONS – DAPSONE
Hypersensitivity to Dapsone and/or its derivatives.

 

10.   DRUG INTERACTIONS – DAPSONE
A modest interaction has been reported for patients receiving 100 mg Dapsone daily in combination with trimethoprim 5 mg/kg q6h. On Day 7, the serum Dapsone levels averaged 2.1 ± 1.0 μg/mL in comparison to 1.5 ± 0.5 μg/mL for Dapsone alone. On Day 7, trimethoprim levels averaged 18.4 ± 5.2 μg/mL in comparison to 12.4 ± 4.5 μg/mL for patients not receiving Dapsone. Thus, there is a mutual interaction between Dapsone and trimethoprim in which each raises the level of the other about 1.5 times.

 

11.     EXCRETION- DAPSONE
Excretion: Urine (85%) as metabolites
